ABSTRACT

Mycoplasma bovis is a highly contagious agent associated with several pathologies in cattle. The detection of reactive antibodies to M. bovis by Indirect Enzyme-Linked Immunosorbent Assay (iELISA) identifies if there was an exposure to the microorganism. The current study aimed to optimize an iELISA from M. bovis total cell antigen, applying it to bovine serum samples, and to evaluate risk factors. Serum samples were obtained from 400 cows from 17 herds from Southeast Brazil. In the optimization of iELISA, the following was established: 2 µg/mL of antigen, sera dilution 1:300, and conjugate dilution 1:15000. The frequency was 62.3% (249/400) of reactive animals and 100% (17/17) of reactive herds. Risk factors were: herds with more than 100 animals (OR= 3.1; CI= 95%); Holstein breed (OR= 72.5; CI= 95%); cows (OR= 29.7; CI= 95%); intensive breeding system (OR= 3.3; CI= 95%); associated small ruminant production (OR= 4.4; CI= 95%); milk production above 500L (OR= 2.9; CI= 95%); no quarantine (OR= 1.5; CI= 95%); mechanical milking (OR= 5.5; CI= 95%) and cases of mastitis (OR= 5.5; CI= 95%). The proposed iELISA was able to detect antibodies reactive to M. bovis in bovine serum. Knowledge of these risk factors can assist in the implementation of prophylactic measures.(AU)


Mycoplasma bovis é um agente altamente contagioso relacionado a várias patologias em bovinos. A detecção de anticorpos reativos a M. bovis por Ensaio de Imunoadsorção Enzimática Indireto (iELISA) identifica se houve exposição ao microrganismo. O presente estudo teve como objetivo otimizar um iELISA de antígeno celular total de M. bovis, aplicando-o a amostras de soro bovino, bem como avaliar fatores de risco. Amostras de soro foram obtidas de 400 vacas de 17 rebanhos da Região Sudeste do Brasil. Na otimização do iELISA foram obtidos: 2µg/mL de antígeno, diluição dos soros 1:300 e do conjugado 1:15000. A frequência de animais reativos foi de 62,3% (249/400) e de 100% (17/17) para os rebanhos. Os fatores de risco foram: rebanhos com mais de 100 animais (OR= 3,1; IC= 95%); raça Holandesa (OR= 72,5;IC= 95%); vacas (OR= 29,7;IC= 95%); sistema intensivo (OR= 3,3; C= 95%); produção de pequenos ruminantes (OR= 4,4;IC=95%); produção de leite acima de 500L (OR= 2,9;IC= 95%); sem quarentena (OR= 1,5;IC= 95%); ordenha mecânica (OR= 5,5;IC= 95%) e casos de mastite (OR= 5,5;IC= 95%). O iELISA proposto foi capaz de detectar anticorpos reativos a M. bovis no soro bovino. O conhecimento desses fatores de risco pode auxiliar na implementação de medidas profiláticas.(AU)

ABSTRACT

There is increasing evidence that neurofilament light chain (NF-L) can be considered as a biomarker for neuro-axonal damage. This polypeptide can be released into the cerebrospinal fluid (CSF) and the blood, where it can be quantified. The concentration of NF-L is elevated in patients with multiple sclerosis (MS) and psychiatric disorders. We aimed to investigate the NF-L levels in the CSF from treated MS patients and the relationship with depression or anxiety. The study involved three groups: control group (individuals without inflammation), the relapse-remitting multiple sclerosis (RRMS)-untreated group, and the RRMS-Fingo group (RRMS patients who were treated with fingolimod). MS disability was assessed by the Expanded Disability Status Scale, and depression and anxiety were evaluated by a neuropsychologist, using the Hospital Anxiety and Depression Scale, the Beck Depression Inventory-II, and the Beck Anxiety Inventory. Individual CSF samples were collected to measure NF-L levels. The results of the statistical analysis on levels of NF-L in the CSF of control subjects, RRMS-untreated patients, and RRMS-Fingo patients were significant. The relationship between depression and anxiety in RRMS-Fingo patients and NF-L levels was not statistically significant. In conclusion, MS events such as anxiety and depression appear to contribute to the onset of clinical relapses, subclinical cases, and neurodegeneration.

ABSTRACT

O objetivo deste trabalho foi estudar a prevalência de MG e MS e a filogenia das cepas circulantes, comparando-as com outras já descritas em poedeiras comerciais no Brasil. Foram coletados 140 suabes traqueais de poedeiras comerciais com sinais respiratórios em seis granjas da região centro-oeste de São Paulo. As amostras foram avaliadas por PCR, com posterior sequenciamento e análise filogenética das cepas identificadas. Das 140 amostras, 16,4% foram positivas para MG e 68,6% para MS. Houve diferença significativa nas frequências de MG e MS por granja, segundo o teste G de independência (P<0,05). Todas as cepas identificadas de MG e MS de granjas distintas apresentaram similaridade tanto pela lipoproteína para MG quanto pela região 16s rRNA para MS. Neste estudo, foi possível observar altas prevalências dos agentes estudados, sendo a de MS maior que a de MG. Foi detectada infecção mista por MG e MS em 11,4% das amostras e sabe-se que esses micoplasmas podem agir de forma sinérgica, agravando o quadro respiratório. As cepas circulantes identificadas, pela análise das regiões gênicas da lipoproteína para MG e 16S rRNA para MS, são similares em todas as granjas estudadas.(AU)


The aim of this study was to evaluate the prevalence of MG and MS and the phylogeny of the circulating strains, comparing them with others already described in commercial laying hens from Brazil. A total of 140 tracheal swabs were collected from commercial laying hens with respiratory signs in six farms from the western region of São Paulo state. The samples were analyzed by PCR with subsequent sequencing and phylogenetic analysis of the identified strains. From the 140 samples, 68.6% were positive for MS and 16.4% for MG. There was a significant difference in the frequencies of MG and MS per farm according to G Test of independence (P<0.05). All strains identified as MG and MS from distinct farms presented similarity both by lipoprotein to MG and by 16s rRNA region to MS. In this study, it was possible to observe a high prevalence of MS compared to MG. Mixed MG and MS infection was detected in 11.4% of the samples. These mycoplasmas may act synergistically, worsening the respiratory signs. The circulating strains identified by analysis of the lipoprotein for MG and 16S rRNA for MS are similar on all poultry farms studied.(AU)

ABSTRACT

O objetivo deste trabalho consistiu na aplicação de indicadores socioambientais em pisciculturas familiares localizadas na região do Vale do Ribeira, em área de Mata Atlântica, no sudoeste do estado de São Paulo. Para isso, foram identificadas 84 propriedades. Dessas, 40 foram selecionadas como unidades amostrais. Entre as principais características identificadas, 32% possuem lâmina d'água inferior a cinco hectares, com viveiros escavados em sistema semi-intensivo, 58% utilizam mão de obra familiar e 23% apontam a ausência de assistência técnica especializada como o principal problema enfrentado. A tilápia-do-nilo (Oreochromis niloticus) representa 57% das espécies produzidas em sistemas de monocultivo ou policultivo. Contudo, as pisciculturas familiares são classificadas como sistemas produtivos de pequeno porte, com destaque para a necessidade de adequação de recursos naturais, da capacidade de gestão e da eficiência das práticas de produção. Os indicadores sociais variaram de 0,75 a 1,00 para equidade salarial, proporção de autoemprego, uso de mão de obra local, inclusão de gênero e inclusão etária, demonstrando a capacidade que a aquicultura tem para auxiliar no desenvolvimento social local, por meio da geração de emprego e renda.(AU)


The objective of this work was to apply socioenvironmental indicators in family fish farms located in the Ribeira Valley region, Atlantic Forest area, southwest of São Paulo state, Brazil. Thus, we identified 84 production units. Of these, we selected 40 as sample units. Among the main characteristics identified, 32% have water depth of less than five hectares with excavated ponds in semi-intensive system, 58% use family labor and 23% report the lack of specialized technical assistance as the main problem faced. The Nile tilapia (Oreochromis niloticus) represents 57% of the species produced in monoculture or polyculture systems. Therefore, family fish farms are classified as small production systems with emphasis on the need to adapt natural resources, management capacity and efficiency of production practices. Social indicators ranged from 0.75 to 1.00 for salary equity, proportion of self-employment, use of local labor, gender inclusion and age inclusion, demonstrating the ability of aquaculture to assist local social development through job and income generation.(AU)

ABSTRACT

Visceral leishmaniasis is a zoonosis in which the dog appears as the main source of infection in urban areas. Its diagnosis is complex and the cytopathological exam is a fast and cheap alternative to parasite direct visualization and its sensitivity can be increased by immunocytochemistry, though with a higher cost. The accuracy of such methods is dependent on the microscopist's experience and therefore, this study evaluated the reliability of such techniques between two observers, from bone marrow aspirates of 50 dogs from an endemic area for the disease. The parasitological culture in Novy-MacNeal-Nicolle medium was used as the reference standard. Among the main findings, the sensitivities obtained by observers I and II were respectively 62.5% and 37.5%, while specificities were 81.1% and 100%. On immunocytochemistry evaluation, the sensitivity was 0% for both evaluators and the specificity 97.3% and 100%. The agreement between evaluators was weak (κ = 0.167) for the cytopathological test and it could not be evaluated for immunocytochemistry, for which there was no detection by the evaluator II. The agreements among the diagnostic methods and the standard reference for the observer I were reasonable (κ = 0.364) for cytopathological examination and bad (κ = -0.041) for immunocytochemistry. For observer II, such agreement could be assessed only for the cytopathological test, being moderate (κ = 0.497). The results point to the possible expertise difference between evaluators, with the evaluator II demonstrating greater experience when interpreting the citopathological test. Although there was the expected sensitivity increase with immunocytochemistry, the technique used in this study was not effective for the diagnosis of infection, regardless of the evaluator.(AU)

ABSTRACT

The presence of the single nucleotide polymorphisms in exon 1 of the mannose-binding lectin 2 (MBL2) gene was evaluated in a sample of 159 patients undergoing coronary artery bypass surgery (71 patients undergoing valve replacement surgery and 300 control subjects) to investigate a possible association between polymorphisms and heart disease with Chlamydia infection. The identification of the alleles B and D was performed using real time polymerase chain reaction (PCR) and of the allele C was accomplished through PCR assays followed by digestion with the restriction enzyme. The comparative analysis of allelic and genotypic frequencies between the three groups did not reveal any significant difference, even when related to previous Chlamydia infection. Variations in the MBL plasma levels were influenced by the presence of polymorphisms, being significantly higher in the group of cardiac patients, but without representing a risk for the disease. The results showed that despite MBL2 gene polymorphisms being associated with the protein plasma levels, the polymorphisms were not enough to predict the development of heart disease, regardless of infection with both species of Chlamydia.

ABSTRACT

A piometra é uma infecção aguda ou crônica do útero que ocorre frequentemente em cadelas não castradas, podendo também ocorrer em gatas domésticas e selvagens, sendo poucos os estudos relacionados à piometra em grandes felídeos. O objetivo deste relato foi descrever um caso de piometra em uma leoa (Panthera leo) de cativeiro, as lesões de necropsia e histológicas, bem como os resultados da análise microbiológica. Uma leoa com aproximadamente 23 anos, pertencente a um criadouro conservacionista de Santa Maria-RS, foi encontrada morta pela manhã em seu recinto. Após coleta de dados, procedeu-se à necropsia e à coleta de material para análise histopatológica e bacteriológica. A análise microbiológica revelou predomínio das bactérias Streptococcus sp. e Escherichia coli no conteúdo purulento do útero, caracterizando como piometra, e a bactéria predominante em plasma, fígado e medula óssea foi E. coli. De acordo com o laudo histopatológico, as alterações observadas nessa leoa sugerem um quadro de septicemia grave, sendo a origem do foco infeccioso bacteriano, provavelmente, a piometra. Considera-se importante chamar a atenção dos médicos veterinários de animais selvagens para um diagnóstico precoce dessa doença, que é comum em cadelas, mas que pode acometer também felídeos selvagens e levá-los à morte.


Pyometra is an acute or chronic uterus infection that occurs often in not spayed dogs, but may also occur in wild and domestic cats, and there are few studies related to pyometra in big cats. The aim of this report was to describe a case of pyometra in a lion (Panthera leo) in captivity, the gross and microscopic lesions found at necropsy, and the results of the microbiological analysis. A female with approximately 23 years of age, belonging to a Conservationist Breeding Center located in Santa Maria-RS-Brazil was found dead in her enclosure in the morning. After data collection, we proceeded to the autopsy and collection of material for histopathological and bacteriological analyzes. Microbiological analysis revealed a predominance of the bacteria Streptococcus sp and Escherichia coli in the uterus content, characterized as pyometra and the predominant bacterium in the plasma, liver and bone marrow was Escherichia coli. According to histopathology, the changes observed in this lioness suggest a framework of severe septicemia, being the source of the bacterial infection, probably, pyometra. It is considered important to draw the attention of zoo and wildlife veterinarians for an early diagnosis of this common disease in dogs, which can also affect large wild cats and lead them to death.

ABSTRACT

Several types of tumors affect dogs' skin. Simultaneously occurring neoplasms with different histological patterns might be rarely present in the same animal. This paper describes the occurrence of epitheliotropic cutaneous T-cell lymphoma and melanoma in a dog. The animal had nodular lesions in the abdominal region and serpiginous plaques on the dorsal region of the trunk. Cytology evidenced malignant fusiform cells from the abdominal lesions as well as few round cells from the dorsal. The histopathological examination of the abdominal lesions showed dermis with polygonal to spindle-shaped neoplastic cells. The lesion of the dorsal region evidenced neoplastic round cells with generally distinct cell borders and a moderate amount of eosinophilic cytoplasm. Abdominal lesions were positive for Melan A. Dorsal and forelimb lesions were positive for CD3. This study reports the occurrence of epitheliotropic cutaneous T-cell lymphoma and malignant melanoma in a crossbred Boxer dog and discusses the importance of performing immunohistochemical profile to confirm the phenotype of the tumor.


Diferentes tipos de tumores podem ocorrer na pele de cães. É rara, porém, a ocorrência simultânea de neoplasias com origens histológicas diferentes no mesmo animal. Este trabalho descreve a ocorrência de linfoma cutâneo epiteliotrópico de células T e melanoma em um cão. O animal apresentava lesões nodulares na região abdominal e placas serpiginosas na região dorsal do tronco e membros. A citologia evidenciou células fusiformes malignas das lesões abdominais, bem como algumas células redondas nas dorsais. O exame histopatológico das lesões abdominais mostrou derme com células neoplásicas poligonais a fusiformes. A lesão da região dorsal evidenciou células redondas neoplásicas com citoplasma eosinofílico. Lesões abdominais foram positivas para Melan A. Lesões dorsais e de membros anteriores foram positivas para CD3. Este estudo relata a ocorrência de linfoma cutâneo de células T epitheliotropic e melanoma maligno em um cachorro Boxer, e discute a importância da realização de perfil imuno-histoquímico para confirmar o fenótipo do tumor. A importância do perfil imuno-histoquímico para confirmar o tipo de neoplasia também é discutida.

ABSTRACT

With the purpose of shedding light on some doubts in veterinary oncology, the present article intends to compare the results of histopathological and immunohistochemical examinations of unspecific round cell neoplasia, to realize immunophenotyping of canine lymphoma cases, to establish the T or B origin of neoplastic cells, and to determine the degree of proliferation and apoptosis of lymphomas by immunohistochemistry. Of 11 animals presenting immunohistochemical diagnosis of lymphoma, five had been diagnosed as Lymphoma by HE staining of histopathological slides and six had been classified as unspecific round cell neoplasia. All cases submitted to immunohistochemical examination were T-cell lymphomas. There was a positive correlation between cell proliferation and apoptosis. The comparison among histopathological and immunohistochemical results obtained in the cases examined in the present study suggested that immunohistochemistry is essential for the differentiation of round cell neoplasia.


Com o objetivo de sanar algumas dúvidas na área da oncologia veterinária, o presente artigo pretende comparar os resultados dos exames histopatológicos e imuno-histoquímicos de neoplasias de células redondas inespecíficas, realizar imunofenotipagem dos casos de linfoma canino e determinar o grau de proliferação e apoptose de linfomas. Dos 11 animais que apresentaram diagnóstico imuno-histoquímico de linfoma, cinco foram diagnosticados como linfoma por coloração HE das lâminas histopatológicas e seis foram classificados como neoplasia de células redondas inespecíficas. Todos os casos submetidos ao exame imuno-histoquímico foram de linfomas de células-T. Houve uma correlação positiva entre a proliferação celular e apoptose. A comparação entre os resultados histopatológicos e imuno-histoquímicos obtidos nos casos analisados sugeriu que a imuno-histoquímica é essencial para a diferenciação das neoplasias de células redondas.

ABSTRACT

A enrofloxacina é um dos antimicrobianos mais utilizados na avicultura industrial, e a deposição de resíduos em produtos avícolas, como os ovos, são de grande importância para a saúde pública. Na legislação brasileira não existe padronização do período de carência para o seu uso na produção avícola e não há Limite Máximo de Resíduo (LMR) fixado para enrofloxacina em ovos. Neste estudo, foi utilizado o kit de ELISA comercial (Bioo Scientific(r)) e a LC-MS/MS na pesquisa de enrofloxacina em ovos de 30 galinhas tratadas previamente via água de bebida, com 10mg/kg de enrofloxacina, durante cinco dias. Seis ovos foram coletados diariamente e analisados durante o tratamento e após a sua suspensão, durante 15 dias. A deposição de resíduos obteve níveis máximos no quinto dia de tratamento das aves, declinando gradativamente até não ser detectada a partir do nono dia de suspensão do tratamento. Considerando como base o LMR de 100µg/kg fixado pelo Brasil para tecidos comestíveis de aves e pela União Europeia para músculo, gordura e pele, após seis dias de suspensão do tratamento, os níveis de resíduos foram inferiores a esse limite, tendo como médias 37,43µg/kg na LC-MS/MS e 14,731µg/kg no ELISA. Dentro das condições deste estudo, um período de carência de seis dias seria mais adequado para utilização dos ovos para consumo humano. Foram detectados valores de resíduos nos ovos menores no ELISA em relação à LC-MS/MS para a mesma amostra, mas os dois métodos apresentaram concordância estatística entre si. A LC-MS/MS é o teste recomendado pela legislação brasileira para a análise de resíduos em alimentos; entretanto, pelos resultados obtidos, o kit de ELISA utilizado também pode ser aplicado na detecção de resíduos de enrofloxacina em ovos, com as vantagens de rapidez e simplicidade...


Enrofloxacin is one of the most used antibiotics in the poultry industry and the deposition of residues in poultry products, such as eggs, are of great concern to public health. In Brazilian law there is no standard withdrawal period for enrofloxacin in eggs and there is no Maximum Residue Limit (MRL) established for this antimicrobial in eggs. In this study, (Bioo Scientific(r)) commercial ELISA kit and LC-MS/MS were used to investigate enrofloxacin in eggs of 30 hens pretreated via drinking water at 10mg/kg of enrofloxacin for five days. Six eggs were collected daily and analyzed during treatment and after the end of treatment, for 15 days. Residues obtained maximum levels on the fifth day of treatment, declined gradually and were no longer detected from the ninth day to the end of treatment. Based on the MRL of 100mg/kg established for edible tissues of poultry by Brazillian law and for muscle, fat and skin, by the European Union, after six days of treatment withdrawal, the residue levels were below that limit, with the average of 37.43mg/kg in LC-MS/MS and 14.731mg/kg in ELISA. Within the conditions of this study, a withdrawal period of six days would be more appropriate to use the eggs for human consumption. The values obtained by ELISA for residues in eggs were lower than those obtained in LC-MS/MS for the same sample, however both methods showed statistical agreement. LC-MS/MS is the recommended method by Brazilian legislation for analysis of residues in food, however, according to the results the ELISA kit used can also be applied to the detection of enrofloxacin residues in eggs, with the advantages of speed and simplicity...

ABSTRACT

O objetivo do presente trabalho foi acompanhar o desenvolvimento imunológico dos neonatos caninos, a fim de avaliar a imunidade celular pela análise dos leucócitos e linfócitos totais e das subpopulações de linfócitos T (CD4+ e CD8+) pela técnica de citometria de fluxo. Foram utilizados 30 cães neonatos de ambos os sexos, sem raça definida, aos três, 10, 17, 24, 31, 38 e 45 dias de idade. A contagem de leucócitos totais aos 45 dias (11.639±3.574) foi significativamente maior que no terceiro dia de idade (8.740±1.812) (P<0,05); não houve diferença entre a contagem total de linfócitos aos 45 dias em relação ao terceiro dia de idade. Quanto às subpopulações de LT CD4+ e LT CD8+, os percentuais de LT CD4+, aos três dias de idade (24,9±16,8 por cento), foram inferiores quando comparados à média entre o 10°, o 24° e o 31°dia (35,5 por cento), e os de CD8+, ao terceiro dia, menores em relação às médias do 10° e do 31° dia de idade. Pode-se concluir que as subpopulações de LT CD4+ e CD8+ sofrem oscilações durante o desenvolvimento pós-natal, sendo estas crescentes em relação aos níveis obtidos aos três dias de idade. A relação CD4+:CD8+ mostrou superioridade para o primeiro tipo celular, sendo que a maior relação entre CD4+ e CD8+ ocorreu no terceiro dia de idade. The aim of this study was to evaluate the immune development of newborn canines, evaluating the cellular immunity by analysis of leukocyte and lymphocyte totals and sub-population of T lymphocytes (CD4+ and CD8+) by the flow cytometry technique. Thirty neonatal mongrel dogs of both sexes were used, at 3, 10, 17, 24, 31, 38 and 45 days of age. The total leukocyte count to 45 days (11639±3574/µL) was significantly greater than on the third day of age (8740±1812) (p<0.05); There were no differences between the total count of lymphocytes at 45 days in relation to the third day of age. As for the subpopulations of CD4+ and CD8+, CD4+ cell percentages, at three days of age (24.9±16.8) they were inferior when compared to the average between the 10th, 24th and 31st day (35.5), and CD8+ cells, on the third day, were lower compared with averages from the 10th and 31st day of age. It can be concluded that the subpopulations of CD4+ and CD8+ cells undergo oscillations during development, with growing postnatal levels obtained at three days old. ABSTRACT

Potencial evocado auditivo é um método eletrodiagnóstico que permite avaliação funcional das vias auditivas desde a orelha média até o tronco encefálico. O uso desse teste em medicina veterinária é pouco difundido no Brasil. O presente artigo relata o uso do potencial evocado auditivo para detecção de deficiência auditiva unilateral em um gato com síndrome vestibular periférica secundária a otite média.


The brainstem auditory evoked potential is an electrodiagnostic test that allows a functional assessment of the auditory pathways from the middle ear to the brainstem. This test, in veterinary medicine, is not commonly used in Brazil. This paper reports the use of auditory evoked potential for deafness detection in a cat with unilateral peripheral vestibular syndrome secondary to otitis media.

ABSTRACT

Objetivou-se avaliar a influência do nível de suplementação com concentrado sobre o perfil hematológico de ovelhas Morada Nova no terço final da gestação e puerpério, bem como comparar os valores obtidos em diferentes categorias (fêmeas em gestação, puerpério e cordeiros). Utilizaram-se 11 ovelhas, aos 75 últimos dias de gestação e nos primeiros 75 dias de lactação, e oito cordeiros progênie das ovelhas, distribuídas em delineamento inteiramente casualizado em arranjo de parcelas subdivididas no tempo. Durante o período experimental, os animais permaneceram em pastagem de Andropogon gayanus, sendo recolhidos ao final da tarde, quando recebiam suplementação concentrada com 19,17% de proteína bruta. As amostras de sangue foram colhidas desde o 75º dia de gestação até o desmame, aos 75 dias de lactação, a cada quatorze dias. O sangue foi colhido sempre pela manhã, antes de os animais serem liberados para o pasto, por punção da veia jugular, para a realização do hemograma. Os diferentes níveis de suplementação não influenciaram (P>0,05) o perfil eritrocitário e leucocitário das ovelhas. A categoria animal influenciou (P<0,05) o teor de hemoglobina e o número de eritrócitos. Os valores médios de hemoglobina não difeririam (P>0,05) entre fêmeas gestantes (7,74g/dL) e puerperais (7,67g/dL), bem como a contagem total de hemácias dos cordeiros (8,14x10(6)/µL) e das fêmeas em puerpério (9,16 x 10(6)/µL). Os valores médios do hematócrito, volume corpuscular médio, concentração de hemoglobina corpuscular média e proteína total não diferiram estatisticamente (P>0,05) entre as ovelhas; no entanto, observaram-se maiores níveis para os cordeiros. O número de leucócitos totais foram superiores (P<0,05) nas ovelhas gestantes e puerperais em relação aos cordeiros. A suplementação concentrada não influenciou o perfil hematológico dos animais, porém a categoria animal exerceu influência sobre os parâmetros hematológicos eritrocitários e leucocitários.


This research was carried out to evaluate the influence of supplementation with concentrate on hematological profile of Morada Nova sheep in last third of pregnancy and postpartum, as well as comparing the values obtained in different categories (pregnant females, postpartum, and lambs). Eleven ewes it the last 75 days of pregnancy and the first 75 days of lactation and eight lambs born from the ewes allotted in completely randomized design and split-plot arrangement in time were used. During the experimental period, the animals remained in Andropogon gayanus pasture. Late in the afternoon they returned to the center of management to receive the concentrate supplementation with 19.17% of crud protein. Blood samples were collected from the 75th day of pregnancy to weaning at 75 days of lactation, every fourteen days. Blood samples were taken always in the morning, before the animals were released into the pasture, by puncturing the jugular vein, for complete blood count. Neither the erythrocyte profile nor the leukocyte profile was affected by the different levels of supplementation (P>0.05). The animal category had a significant effect (P<0.05) on hemoglobin and erythrocyte number. The mean hemoglobin values did not differ (P>0.05) between pregnant ewes (7.74 g/dL) and ones in postpartum (7.67g/dL) as well as the total count of red blood cells of the lambs (8.14 x 106/µL) did not differ (P>0.05) from females in postpartum (9.16 x 106/µl). When comparing between pregnant and postpartum ewes for the mean hematocrit values, mean corpuscular volume, mean corpuscular hemoglobin concentration and total protein did not differ (P>0.05). The highest values were observed in these animals. The total white cell count was higher (P<0.05) in pregnant and postpartum ewes than lambs. The concentrate supplementation did not affect the blood profile of animals, but the animal category exerted influence on erythrocyte and leukocyte hematological parameters.

ABSTRACT

Collibacillosis is considered one of the major diseases of the modern poultry industry, due to the significant losses it causes. Escherichia coli contributes not only to the disease itself, by causing weight loss of the birds, but also to the increase in carcasses condemnation during slaughter and processing. Detection of virulence factors in E. coli strains of the APEC pathotype contributes to the characterization and pathogenicity of this agent. PCR techniques have been very helpful in the search for genes that encode those virulence factors. This study aimed to detect the gene Fel A of E. coli by PCR and relate its positivity to low weight in broiler flocks with airsacculitis as diagnosed by the health inspection service. The study involved 40 flocks of broilers slaughtered in a single poultry slaughterhouse, under Federal Sanitary Inspection, located in the state of Rio Grande do Sul, Brazil. Three broilers were randomly selected to obtain one "pool" of three tracheas for each PCR. DNA was extracted using phenol-chloroform and amplified using a pair of primers specific to gene Fel A of E. coli. Of the 40 flocks analyzed by PCR, 35% (14/40) were positive for the gene Fel A. PCR was an effective technique for the detection of gene Fel A in broiler flocks. A colibacilose é considerada uma das principais doenças da indústria avícola moderna, devido aos grandes prejuízos econômicos causados. A Escherichia coli contribui não só para a doença em si, levando à perda de peso das aves, bem como para o aumento da taxa condenação de carcaças durante o abate e processamento. A detecção de fatores de virulência de cepas de E. coli do patotipo APEC colabora para a caracterização de sua patogenicidade e as técnicas de PCR têm sido muito úteis na pesquisa de genes que os codificam. Este estudo objetivou diagnosticar E. coli pela detecção o gene Fel A por PCR e relacionar a positividade para este agente com o baixo peso em frangos de corte provenientes de lotes condenados por aerossaculite. Foram estudados 40 lotes de frangos de corte abatidos em um matadouro avícola sob Inspeção Sanitária Federal, localizado no Estado do Rio Grande do Sul. Foram colhidos aleatoriamente 3 frangos e obtidos "pools" de três traqueias em cada um deles para PCR. O DNA foi extraído pelo método de fenol-clorofórmio e amplificado com pares de "primers" específicos para gene Fel A de E. coli. Dos 40 lotes analisados pela PCR, 35% (14/40) foram positivos para o gene Fel A.
